I'm hoping that this riddle will be a bit more challenging than usual, but I have been proven wrong before.
I challenge you, guardians, to search your minds for the absolute limit and uncover the answer.
This may be easy, or it may be hard. The choice is yours ;)

Who/what am I?

I am the third, brought into existence when the hallowed crown fell before the undead warrior. I brought about something no one ever saw before, and no one will ever see again. I can be made stronger, and in this strength, I can provide insight into my home. You will know me, for the truth you are about to see, will mark you forever.[/i]"

The Vault of Glass location (referred to extremely vaguely with the word "time") has been used three times for a specialized mission.
"Once, it harbored the architects of our suffering, weaving and unweaving the fabric of existence as we trudged to reach the pinnacles of a now-ancient glory."
The first time it was used was for the Vault of Glass raid, which was the first ever raid in Destiny. The architects of our suffering are the vex, which control existence inside of the vault of glass. The "now-ancient glory" was the coveted Level 30, which could only be achieved through raid gear at the time.
"The second, overrun by the disciples of the possessed ruler, clinging to the hope that destruction will be their retribution."
The second time the Vault of Glass is used is in the mission Wolves' Gambit in HoW. The disciples of a possessed ruler are the followers of Skolas. And Skolas could be considered "possessed" based on the theory that the horns on Skolas' back are Ahamkara horns that are influencing him to be vicious and tyrannical. They raid the Vault of Glass in hopes of finding weapons that will redeem the House of Wolves name and bring new glory to the fallen.

The third time VoG was used was in the Paradox story mission in TTK. The mission is unlocked after your guardian (the undead/resurrected warrior) defeats Oryx (the hallowed crown) in the Regicide mission. In the Paradox mission, you fight a unique creature, a taken Hydra, which can only be found in that mission as far as I know.
"I can be made stronger, and in this strength, i can provide insight into my home."
This line refers to the Heroic version of the mission, where the level increases to 41, and there are hidden secrets in the heroic version that reward grimoire (insight) for the vault of glass.
"You will know me, for the truth you are about to see, will mark you forever."
The Paradox mission isn't exactly required, but it is one of the main vanguard missions after the Regicide missions. The "truth" that you learn is the story of Praedyth, and the fact that he is now dead. The Vision of Confluence description also served as a reference to Vault of Glass.[/spoiler]
