Assuming you are running D2 on Steam, just have your brother make his own Steam account and then proceed to make a new D2 account

One key item to note with running more than one Steam account and Destiny 2. Destiny 2 is not a title that can be shared using Steam's Family Library Share feature. This means that each Steam account has to purchase each expansion separately even when playing from the same computer.

Yeah and also i'm sure you're probably aware that a new account means your brother will have to purchase any expansions himself. He won't share yours
