Hardest in recent memory would have to be Marie 300%. Still haven't beat her.
Also trying to beat Ninja Gaiden II on Mentor difficulty, I'm going to say all of the bosses.

Can't remember on Mentor, but Dagra Dai on Master Ninja is by far the hardest boss in the game. Which is odd considering he was a pushover on lower difficulties. Good luck.
