Throughout the course of playing many.... many times through Devil's Lair in both the Alpha and the Beta, several of us from ENIGMAS began to think about the 6 man raids. It occurred to us that there was no really good way of designating targets as a focus to be taken down by the team.
For instance, we might find ourselves saying take down the captain first. Well, then the question might become "which captain?" To which our only reasonable reply might be "the one with 45% shields left, not the one with 55%" which leaves many people in a situation where they have to stop and really stare at each of the targets, which may be on opposite sides of the battlefield in order to determine which target is the one that they should be hitting.
This is of course assuming that the enemies in the yet to come raids are strong enough to resist 1 to 2 people's dps for a sustained period. Fortunately, an easy solution to this issue became readily apparent upon consideration. We noticed that there was no functionality bound to the directional swipe of the touchpad on the Dualshock 4 controller and thought to ourselves, that this would be the easiest way to implement a marking system.
In example, you might be able to map 4 marks (i.e. skull, circle, square and triangle) to the up, down, left and right Swipes on the touch pad.
That being said, I have no idea what you might be able to do for those people who play on the XBox consoles.... But their lack of controller functionality is no reason to limit that of the PS4. Perhaps the brilliant mind @Bungie will be able to figure out a better way to handle it for the other controller types.

cool idea, that would be really helpfull for players without voice chat. PS4 touchpad would be something easy to use.

I wonder if this something that would make sense to bind to R3 so if you are targeting a player it will select them if you are targeting an enemy it will mark them for everyone in your fireteam.

I do agree with targeting helpers, maybe more so with Devil Walkers and such other enemies that have x number of weak spots. (FYI that Walker needs to drop loot for how long it takes to kill it (>_<) )

This would make raids considerably easier, if what they say about the difficulty level is true. And since there is no functionality for the touchpad at the moment, this would be a perfect idea.

There has to be a solution that works across systems. But marking would add to the group co-op feel this game is trying to achieve.
